Numerous eukaryotic DNA processing enzymes, such as DNA polymerases and ligases, bind the processivity factor PCNA, which acts as a platform to recruit and regulate the binding of enzymes to their DNA substrate. Multiple PCNA‐interacting motifs (PIPs) are present in these enzymes, but their individual structural and functional role has been a matter of debate. Recent cryo‐EM reconstructions of high‐fidelity DNA polymerase Pol δ (Pol δ), translesion synthesis DNA polymerase κ (Pol κ) and Ligase 1 (Lig1) bound to a DNA (...) substrate and PCNA demonstrate that the critical interaction with PCNA involves the internal PIP proximal to the catalytic domain. The ancillary PIPs, located in long disordered regions, are instead invisible in the reconstructions, and appear to function as flexible tethers when the enzymes fall off the DNA. In this review, we discuss the recent structural advancements and propose a functional hierarchy for the PIPs in Pol δ, Pol κ, and Lig1. (shrink)

The literary interior, carrying out the most important function in the text of any literary work, plays a key role in revealing the author’s intention and in the narration strategy. Unlike the landscape and nature description, the interior is a depiction of the “closed space” where the protagonist lives. This space keeps traces of the hero’s habits, psychological state, and other features. As the literary-aesthetic trends replaced each other, the interior’s variety also changed, and over the time, it attained literary (...) functions namely in the establishment of the text and in revealing the author’s idea and intention. The description of any space in the text of a literary work is not casual, as after this description we are witness to the depiction of hero living in this space; the matter is that these two successive depictions are not forgotten and stimulate the revealing of various significance levels occurring in the literary text. There are explicit and implicit interiors. The successive description of the space in the literary text is of symbolic meaning and serves to reveal its functionality. If, say, a carpet is described in the space of an artistic work, or its existence is confirmed in this space, it is necessarily related to the meanings hidden in the inner world of the hero. Cancers rely on multiple, heterogeneous processes at different scales, pertaining to many biomedical fields. Therefore, understanding cancer is necessarily an interdisciplinary task that requires placing specialised experimental and clinical research into a broader conceptual, theoretical, and methodological framework. Without such a framework, oncology will collect piecemeal results, with scant dialogue between the different scientific communities studying cancer. We argue that one important way forward in service of a more successful dialogue is through greater integration of applied sciences (experimental and clinical) (...) with conceptual and theoretical approaches, informed by philosophical methods. By way of illustration, we explore six central themes: (i) the role of mutations in cancer; (ii) the clonal evolution of cancer cells; (iii) the relationship between cancer and multicellularity; (iv) the tumour microenvironment; (v) the immune system; and (vi) stem cells. In each case, we examine open questions in the scientific literature through a philosophical methodology and show the benefit of such a synergy for the scientific and medical understanding of cancer. BackgroundA different ethos with respect to the perception of medical ethics prevails in societies in transition such as those in the Arabian Peninsula, which makes it difficult to apply international principles of bioethics in medical practice. This study aimed to develop and psychometrically test an instrument that measures physicians’ awareness of bioethics and medical law and their attitudes towards the practice of medical ethics. Additionally, it examined physician correlates influencing the awareness of bioethics.MethodsFollowing a rigorous review of relevant literature by (...) a panel of experts, a 13-item instrument, the Omani physicians’ bioethics and medical law awareness (OBMLA) questionnaire was developed with the aim of assessing physicians’ awareness of bioethics and medical law. The study tool’s construct validity and internal consistency reliability were examined by exploratory factor analysis (EFA) and Cronbach’s alpha. In a cross-sectional study, the questionnaire was distributed among a random sample of 200 physicians at a tertiary hospital in Muscat, Oman. Participant characteristics that may influence awareness of bioethics and medical law were explored.ResultsThe EFA of the OBMLA questionnaire resulted in three well-loading factors: (1) Physicians’ bioethics practice subscale (2) incentive related bioethics subscale and (3) medical law awareness subscale. Internal consistency reliability ranged between Cronbach’s α: 0.73–0.8. Of the total 200 participants, 52% reported that teaching medical ethics during medical school was inadequate. The overall mean (standard deviation, SD) of the bioethics awareness score and Omani medical law awareness were 27.6 (3.5) and 10.1 (2.1) respectively. The majority of physicians (73%) reported that they frequently encountered ethical dilemmas in their practice and 24.5% endorsed the view that unethical decisions tended to occur in their practice.ConclusionThe study provides an insight into the practice of bioethics, and the awareness of bioethics and medical law among physicians in a teaching hospital in Oman. The OBMLA questionnaire appears to be a valid and reliable tool to assess a physician’s awareness of bioethics and medical law. In this preliminary study, it appears that participants have suboptimal scores on the indices which measure practice and awareness of bioethics and medical law. (shrink)

It's surprising that contemporary moral philosophers have not thought more about food. The rapidly expanding industrialized landscape of modern western agribusiness raises moral concerns about large-scale livestock production, the increased usage of genetically modified crops, and the effects these now common practices may have on long-term environmental and human health. Here Pence argues that biotechnology is more helpful than harmful, on the ground that it will abate world hunger. His argu- ment is simple: the claim that GM foods are unsafe is the product of alarmism, not sound reason. Dès les années 1940, le philosophe Rudolf Carnap s'est proposé de relever le «défi humien» en fondant une théorie de la confirmation par la construction d'une logique probabiliste dite «inductive». Ce projet avait été esquissé à Cambridge dans les années 1920 par l'économiste John M. Keynes. Examiner le programme de Carnap dans sa totalité, le situer (...) dans son contexte historique, suivre son évolution et les influences qu'il a eues, tels sont les buts de cet article. The idea of associating probability and induction is not a theme suitable for the xxth century but it has received a systematic development when the neo-positivist philosophers seized it. Beginning with the 1940's, the philosopher Rudolph Carnap proposed to affront the «Humian challenge» by founding a theory of confirmation through the construction of a probabilistic logic called «inductive». This project had been outlined in Cambridge during the 1920's by the economist John M. Keynes. Because of his preoccupation with the formal aspects of music and literature, Theodor W. Adorno is often regarded as the most aesthetically oriented thinker of the Frankfurt School theorists. It is Adorno’s perceived commitment to aestheticism—the study of art for art’s sake and the study of art as a source of sensuous pleasure, rather than as a vehicle for culturally constructed morality or meaning—that many scholars have criticized as hostile to genuine, concrete, substantive political, social, and ethical engagement with the (...) arts. _Adorno and Ethics_—the first issue of _New German Critique_ to be published by Duke University Press—takes issue with Adorno’s critics. Many philosophers agree that Hume was not simply objecting to inductive inferences on the grounds of their logical invalidity and that his description of our inductive behaviour was inadequate, but none the less regard his argument against induction as irrefutable. I argue that this constellation of opinions contains a serious tension. In the light of the tension, I re-examine Hume’s actual sceptical argument and show that the argument as it stands is valid but unsound. I argue that it can only (...) be converted into a sound one if our inductive behaviour can be characterized as a process of rule-governed ampliation. Drawing on some Bayesian ideas, I argue that our inductive behaviour probably cannot be characterized in that way, so our immunity from Hume is secure. Finally, I compare my response to Hume’s argument with some other well known responses. (shrink)

The study aimed to identify the strategic sensitivity and its impact on enhancing the creative behavior of Palestinian NGOs in Gaza Strip, and the study used the descriptive analytical approach and the questionnaire as a main tool for collecting data from employees of associations working in Gaza Strip governorates, and the cluster sample method was used and the sample size reached (343) individuals (298) questionnaires were retrieved, and the following results were reached: The relative weight of strategic sensitivity was 79.22 (...) (%), and the relative weight of creative behavior was 78.99 (%), a statistically significant relationship between all strategic sensitivity and creative behavior, and the presence of a sensitivity effect The strategy’s strategy on creative behavior, there are statistically significant differences in the scale dimensions attributable to the gender variable and the differences were in favor of females, there are no statistically significant differences between the averages of strategic sensitivity due to the age variable, and the educational qualification, and there were no statistically significant differences in creative behavior according to The gender variable, age, educational qualification, specialization, and the study presented a set of recommendations, the most important of which are: the need for civil organizations in Gaza Strip to seek funding from external countries in order to provide self-income for associations to face crises and give them independence Mechanism in order to keep them to carry out their role in society, the need to follow up the strategic plan of civil organizations using e-mails as they pave the way to reach excellence and creativity in the field of work. I examine the argument that scientific theories are typically 'underdetermined' by the data, an argument which has often been used to combat scientific realism. I deal with two objections to the underdetermination argument: (i) that the argument conflicts with the holistic nature of confirmation, and (ii) that the argument rests on an untenable theory/data dualism. I discuss possible responses to both objections, and argue that in both cases the proponent of underdetermination can respond in ways which are individually plausible, but (...) that the best response to the first objection conflicts with the best response to the second. Consequently underdetermination poses less of a problem for scientific realism than has often been thought. (shrink)

Hamilton’s theory of inclusive fitness is a widely used framework for studying the evolution of social behavior, but controversy surrounds its status. Hamilton originally derived his famous rb > c rule for the spread of a social gene by assuming additivity of costs and benefits. However, it has recently been argued that the additivity assumption can be dispensed with, so long as the −c and b terms are suitably defined, as partial regression coefficients. Resilience may be referred to as the capacity for positive adaptation and to quickly recover from difficulties and significant adversity. After examining operational definitions of related concepts, the article discusses resilience building exercises for functional fitness at the individual soldier level, to include among others, self-monitoring, self-evaluation, self-reinforcement, emotional regulation exercises, mindfulness training, relaxation and grounding exercises and importance of maintaining discipline and routine in the military. Using an acronym CARRIES, the article examines efforts to enhance resilience building through empirically (...) validated cognitive, behavioural and intrapersonal skills originating from best practices learned in the military. Architectural theory arises from building, when the mind considers its symbolic relations to its own constructions. The intent of this essay is to discuss the intellectual causes that precede building and precede theory. It considers certain fundamental dualities in our thinking about architecture—such as image and word; type and model; imitation and invention—and the role they play in its making, its perfection as an art, and the eventual elaboration of its tenets into a theory. At a time when theories of (...) architecture proliferate as expressions of ‘personal philosophies,’ a careful and incisive philosophical approach to if, how, and when does theory become formative of building, may ensure that architecture remain faithful to its intrinsic purposes. Hox proteins are part of the conserved superfamily of homeodomain‐containing transcription factors and play fundamental roles in shaping animal body plans in development and evolution. However, molecular mechanisms underlying their diverse and specific biological functions remain largely enigmatic. Here, we have analyzed Hox sequences from the main evolutionary branches of the Bilateria group. We have found that four classes of Hox protein signatures exist, which together provide sufficient support to explain how different Hox proteins differ in their control and function. (...) The homeodomain and its surrounding sequences accumulate nearly all signatures, constituting an extended module where most of the information distinguishing Hox proteins is concentrated. Only a small fraction of these signatures has been investigated at the functional level, but these show that approaches relying on Hox protein alterations still have a large potential for deciphering molecular mechanisms of Hox differential control. Contrary to the general belief that modernist art and architecture reflect the technological society, Jacques Ellul maintains in his L’empire du non-sens that they are justifications for the integration of humankind into what he called the technicist complex. Modernism in art and architecture meant that every product must be qualified by a technological character. This unassailable belief exerted some far-reaching influences on symbolic thought, on artistic expression, on architectural character. If imitation and invention were the two inseparable concepts through which (...) art and architecture were produced and symbolized, it is because they operate on a certain transparency between an exemplar and a work of art. By contrast, in the mentality of technique, artistic production operates based on a certain opacity to meanings outside of itself. Imitation and invention were separated. Thus, one of the principal beliefs in modernism, that art and architecture symbolize the technological society is actually one of its weakest. In considering technology as both the symbol and the product, the artistic idea and its representation collapsed into each other. (shrink)

This essay assesses the opposition of pluralism and monism with respect to politics and architecture, developing the argument within three general areas: the spurious association between political intentions and architectural character, the distinctions and commonalties between political freedom and artistic freedom, and the adverse effect of inappropriate associations between political content and artistic form in general and, in particular, the perceptual impairment of the processes by which buildings come to be endowed with their suitable character.

We propose a new relevance sensitive model for representing and revising belief structures, which relies on a notion of partial language splitting and tolerates some amount of inconsistency while retaining classical logic. The model preserves an agent's ability to answer queries in a coherent way using Belnap's four-valued logic. Axioms analogous to the AGM axioms hold for this new model. The distinction between implicit and explicit beliefs is represented and psychologically plausible, computationally tractable procedures for query answering and belief..

Several authors have recently suggested that classic psychedelics might be safe and effective agents of moral enhancement. This raises the question: can we learn anything interesting about the nature of moral experience from a close examination of transformative psychedelic experiences? The interdisciplinary enterprise of philosophical psychopathology attempts to learn about the structure and function of the “ordinary” mind by studying the radically altered mind. By analogy, in this chapter we argue that we can gain knowledge about the everyday moral life (...) by studying extraordinary experiences of altered moral cognition and experience. We focus on a specific class of cases: experiences of forgiveness induced by psychedelics. We argue that close attention to such experiences reveals the importance of thought/emotion coherence and dissonance in the moral life and vindicates some heterodox ideas about moral phenomenology and psychology.
